Proofread it, looking for errors. I've written two novels which I self-published, so I was ultimately entirely responsible for their content. Even after reading them many times, I was never able to discover all the errors, unfortunately. Your goal should be to produce a professional manuscript, one with as few glaring mistakes as possible. If you are successful in selling your story, your publisher will edit it further for you, making suggestions regarding the story-line itself. Thus, you should be predominantly concerned with polishing your manuscript in order to attract a literary agent to represent you, as almost certainly you will require an agent's services, and he/she will be more likely to agree to doing so if your manuscript is well-written and virtually error-free. ask your friends and family to read it and tell them to be honest in their opinions
then ask a third party observer to read it because you're friends and family are most definitely going to lie
go away from it and think about other stuff for a while, then come back and look at it again. something that made sense while you were writting it might not now.
if you have someone whos good with grammer and spelling, then thats always helpful
